Imaging transient cell physiology
Calcium transit across the cell membrane is essential
to nerve impulse transmission, hormone
secretion, and many other essential physiological
functions. Imaging calcium efflux, within a single
cell or simultaneously in multiple cells, requires
high-throughput screening to be most applicable
to normal and aberrant physiology.
In Chapter 4 of this eBook, we review the use of an
automated analysis system for measuring calcium
efflux from bovine aortic endothelial cells. Fluo-8
intracellular fluorescence indicators show calcium
mobilization upon treatment with adenosine
triphosphate. Different cells in the same field of
view exhibit different calcium flux patterns over
a >2 min time period. Thus, one can readily determine
synchronous and asynchronous calcium flux
by a straightforward protocol. CELENA imaging
systems available from Logos Biosystems are also
compatible with trace fluorescence, Z-stack, and
time lapse analysis.
Concluding remarks
Neurology research has often been conducted under
conditions that do not reliably mimic human
physiology, or has focused on one cell at a time in
a manner that hinders physiological interpretation.
These limitations have slowed drug discovery and
development, but are no longer necessary with
modern tools and technologies. In this introduction,
we have provided a brief overview of creating
organoid models that mimic the physiology of the
nervous system, and its applicability to neuroinflammatory
and neurodegenerative diseases. We
have also presented the utility of automated analysis
tools for measuring calcium flux in multiple cells
at a time.
