Sonication is a key method for preparing alphasynuclein pre-formed fibrils (PFFs) for use
in vitro and in vivo. The process is instrumental in
solubilizing the fibrils, which is likely to be a key
element underpinning their activity performance
in downstream experiments, rather than the fibril
length generated.

Additional information
Different monomers and preparation methods
produce different types of fibrillar constructs. They
vary in structure, endotoxin levels, and aggregation
activities. Type 1 PFFs have high seeding capacities
and endotoxin levels between 10-20 EU/mL. Type
2 PFFs have limited seeding capacities and lower
endotoxin levels (<2 EU/mL). Type 3 PFFs have
similar structures and properties as Type 1 PFFs but
lower endotoxin levels (<2 EU/mL). Read more about
