Grunt was set-up and running with only 30 minutes of handson time. Grunt took 12 hours to complete buffer exchange and
concentration. Post exchange, mass recovery and concentration
were analyzed for each formulation. Concentration for the
samples ranged from 55-70 mg/mL and had mass recoveries
of 97-100% (Figure 6).

Conclusion
Automated buffer exchange reduces hands-on time and enables
more precise control over protein volume and quality compared to
manual methods. The Grunt and Junior platforms use a UF/DF,
pressure-based methodology with gentle orbital mixing, coupled
with volume verification to give the operator more direct control
over how their protein is exchanged into a new buffer. Both these
tools delivered on final mass recovery, concentration and stability.
Utilizing this methodology, with Junior for early stage candidate
selection and Grunt for later stage preformulation screening and
final formulation scale-up, can be highly impactful for biologic
drug development.
