Unique background correction
Quantification of turbid, highly concentrated samples can
be troublesome and requires a proper turbidity correction.
Conventional UV/Vis quantification instruments pick a
wavelength (e.g., 340 or 405 nm) where no moleculespecific absorbance is to be expected to perform a background correction. This holds true if the scatter of the light
at each wavelength is the same, which is not the case
(Figure 3). Therefore, on the Lunatic, next to the single wavelength background correction, a more optimal background
correction can be selected applying a turbidity profile taking
wavelength-dependent scatter profiles into account.
Pushing the limits
Biologics concentrations in biopharmaceuticals are increasing,
demanding instruments for reliable, batch wise, fast and
dilution-free quantification. Therefore, the Lunatic platform
has been optimized to increase the upper limit to 275 OD.
